What is the minimum clearance between tyres and frame for a bike being ridden on and off road?

Well, there's a theoretical answer I can give you. Most bikes being sold in the market today meet ISO-4210 standards. The tire clearance requirement to pass ISO-4210 is 6mm. 

The practical answer is a lot less than 6mm but it depends on what tire, tire pressure terrain etc. The best way to find out is to try. If you go the 6mm route you will never have any problems guaranteed.

That's interesting that there is a standard tire clearance.  I have 5mm clearance and I decided that was pretty good.  We'll see if I do any rides with severe mud, then I'll know.

Now, for those worried about their carbon frames, just put helicopter blade (protection) tape in the areas that are vulnerable.  It won't prevent mud build up, but it will protect the fiber structure.

Yeah, Mark is right on the ISO standard.   That is what most companies are going to use to quote maximum tire size.

I have an aluminum bike where my rear tire had about 2mm clearance on both sides.  When I put the torque down, the tire would rub on the frame and actually cut into the chain stay.  Ugh!

Then there is the problem if you break a spoke - you'll want some room for a warped wheel to wiggle around if you want to ride the bike home.  
Mud of course changes everything.  The more clearance the better.

So, 6mm is relatively safe.  2mm is living on the edge.  4mm is doable in most circumstances.  You can decide what you feel comfortable with.
